Summary and explanation TdT is a 58 kDa protein (2) that catalyses the polymerisation of deoxynucleotides on the 3’ hydroxyl end of a polydeoxynucleotide without template instructions (3, 4). TdT is present in the nuclei of normal T and B lymphocyte precursors and the neoplastic equivalent (5). Positive cells are present in normal thymus, and in few normal bone marrow cells, corresponding to hemapoietic precursors (2). Most cases of ALL are TdT positive, 37 % of AML cases are TdT positive (3). Reagent provided F7139, is a purified monoclonal mouse antibody conjugated with Fluorescein Isothiocyanate Isomer 1 (FITC). The conjugate is provided in liquid form in buffer containing 1% bovine serum albumin (BSA) and 15 mmol/L NaN3, pH 7.2. Each vial contains 50 tests (10 µL of conjugate for up to 106 MOLT-4 cells). Application Fluorescent Mounting Medium enhances the visualization of specimens when viewed under a fluorescent microscope. The unique formulation of this product retards the fading of fluorescence allowing the specimens to be reviewed long after mounting. Slides that are mounted with Fluorescent Mounting Medium and stored at 2–8°C in the dark preserve thei r fluorescent signal for at least one month. Fluorescent Mounting Medium dries compley when slides are coverslipped forming a solid coating that facilitates handling and storage. Procedure 1. Dispense one or two drops of Fluorescent Mounting Medium onto tissue sections or cell preparations which are mounted on a glass microscope slide. 2. Apply glass coverslip over Fluorescent Mounting Medium and allow the medium to spread evenly over the specimen. 3. Slides may be examined immediay under a fluorescent microscope. However, the coverslips should not be moved as mounting medium will not compley solidify for several minutes. 4. For long-term storage the edges of the coverslip may be sealed with nail polish to prevent air pockets from forming.

The antibody reacts with the mu-chains of human IgM. Traces of contaminating antibodies have been removed by solid-phase absorption with human plasma proteins.
In crossed immunoelectrophoresis, using 12.5 µL antibody per cm2 gel area against 2 µL human plasma, only one precipitate corresponding to IgM appears. Staining: Coomassie Brilliant Blue.
In indirect ELISA no significant reaction is observed when using human IgG and IgA as coating antigens. In double antibody sandwich ELISA no significant reaction is observed with human plasma stripped of IgM.
Cross-reaction with IgM from other species may occur. As demonstrated by rocket immunoelectrophoresis, the antibody cross-reacts with the IgM-equivalent protein in cat, cow, deer, dog, elephant, hamster, horse, mink, mouse, polecat, raccoon dog, rat, seal, sheep and swine (9).
