| Background Information | Some of the tyrosine residues can be tagged with a phosphate group (phosphorylated) by protein kinases. (In its phosphorylated state, it is referred to as phosphotyrosine.). Tyrosine phosphorylation is considered as one of the key steps in signal transduction and regulation of enzymatic activity. The advent of anti-phosphotyrosine antibodies is one of significant events in signal transduction research. Before the availability of anti-phosphotyrosine antibodies, tyrosyl phospyhorylation of proteins and enzymes was investigated through hazardous and time-consuming radioactive experiments. Anti-phosphotyrosine antibodies are commonly used in western blots after the targeted proteins have been immunoprecipitated to measure the tyrosyl phosphorylation of the proteins. Anti-phosphotyrosine antibodies are also directly used on cell lysate to examine the overall change of tyrosine phosphorylation level in reponse to various treatments. |

BACKGROUND
Focal adhesions were identified as areas within the plasma membrane of
tissue culture cells that adhere tightly to the underlying substrate. In vivo,
these regions are involved in the adhesion of cells to the extracellular matrix.
Paxillin and vinculin are cytoskeletal, focal adhesion proteins that are components
of a protein complex that links the Actin network to the plasma
membrane. Vinculin binding sites have been identified on other cytoskeletal
proteins, including Talin-1 and α-actinin. In addition, vinculin, Talin-1, Talin-2
and α-actinin each contain Actin binding sites. Expression of vinculin, Talin-1
and Talin-2 have been shown to be affected by the level of Actin expression.
α-actinin has been shown to link Actin to integrins in the plasma membrane
through interactions with the vinculin and Talin complex or by a direct interaction
with integrin. Talin-2 is similar to Talin-1 but shows distinct patterns of
expression and cannot compensate for the loss of Talin-1.

Talin-1 （YQ-16） is recommended for detection of Talin-1 of mouse, rat and

（1 ml of cell lysate）], immunofluorescence （starting dilution 1:50, dilution
range 1:50-1:500）, immunohistochemistry （including paraffin-embedded
sections） （starting dilution 1:50, dilution range 1:50-1:500） and solid phase
ELISA （starting dilution 1:30, dilution range 1:30-1:3000）.

Intended use For In Vitro diagnostic use.
This product is intended for use as a diluent in the preparation of primary antibodies and negative control
reagents. It is to be used with antibodies and/or antigens that yield high background in immunohistochemical
（IHC） staining procedures.
Refer to the “General Instructions for Immunohistochemical Staining” or the Detection System “Instructions” of
IHC procedures for: （1） Principle of Procedure, （2） Materials Required, Not Supplied, （3） Storage, （4） Specimen
Preparation, （5） Staining Procedure, （6） Quality Control, （7） Troubleshooting, （8） Interpretation of Staining,
（9） General Limitations.
Reagent provided 0.05 mol/L Tris-HCl buffer containing 0.1% Tween, stabilizing proteins to reduce background, and 0.015 mol/L
sodium azide.
Precautions 1. For professional users.
2. This product contains sodium azide （NaN3）, a chemical highly toxic in pure form. At product concentrations,
though not classified as hazardous, build-ups of NaN3 may react with lead and copper plumbing to form
highly explosive metal azides. Upon disposal, flush with large volumes of water to prevent azide build-up in
plumbing.1,2
3. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin.
4. Unused solution should be disposed of according to local, State and Federal regulations.
Reactivity For demonstration of tissue antigens by IHC, optimal staining performance is dependent on the correct dilution
of the primary antibody. The interaction between antibodies and epitopes consists mainly of relatively weak
forces such as van der Waals forces, electrostatic forces and hydrophobic forces, which are influenced by pH
and ionic strength. Consequently, to assure optimal performance of the antibody with the least amount of
background, the choice of diluent is important.
Applications This product provides an appropriay buffered solution for the dilution of both polyclonal and monoclonal
antibodies and for the preparation of negative control reagents in IHC. This product is particularly useful for
reducing background staining while maintaining specific reactivity. Antibody Diluent with Background Reducing
Components can also be used to stabilize diluted antibodies when stored at 2–8°C. This diluent should be
brought to room temperature and mixed thoroughly prior to use.
Limitations Tissue staining results are dependent upon the handling and processing of the tissue prior to staining.
Aldehyde-containing fixatives, such as formalin and glutaraldehyde frequently contribute to increased
background staining. Improper fixation, freezing, thawing, washing, drying, heating or sectioning may produce
artifacts, antibody trapping or false-negative results. Also, false-positive staining may be caused by crossreactivity
of other IHC staining reagents to e.g. endogenous alkaline phosphatase, endogenous peroxidase,
pseudoperoxidase, endogenous avidin-binding activity and/or by nonspecific reaction with necrotic or
degenerated cells.
Storage Store at 2–8°C. Do not freeze.

Description
Autophagy is a process of bulk protein degradation in which cytoplasmic components, including organelles, are enclosed in double-membrane structures called autophagosomes and delivered to lysosomes or vacuoles for degradation. Atg12 is the human homolog of a yeast protein involved in autophagy （1）. It is essential for Atg12 conjugation, and the ATG12-ATG5 conjugate is necessary to form autophagic vesicles （2）. The E3-like activity of the Atg12-Atg5 conjugate potentiates the formation of Atg8-PE; making the autophagic pathway analogous to ubiquitin-like pathways where an E3 conjugation ligase is required （3）.

Application & Usage: Western blotting （0.5-2 µg/ml）. However, the optimal concentrations should be determined individually. Other applications have not been tested. In mouse intestine tissue lysate, the antibody recognizes a ~100 kDa band （possibly dimer）. Reactivity to other species has not been tested.

Background Descriptions: Beclin is a Bcl-2 binding protein. Its structure includes a Bcl-2 binding coiled-coil region, and a leucine-rich nuclear export signal （NES）. Beclin protein colocalizes with intracytoplasmic organelles and nuclei in normal COS7 and MCF7 cells. Beclin interaction with Bcl-2 may be involved with host viral defense, since over expression of beclin inhibits Sindbis virus replication and expression of beclin lacking the Bcl-2 binding domain has no antiviral effects.
Handling: The antibody solution should be gently mixed before use.

8-Nitroguanosineは、一酸化窒素（NO）と活性酸素（スーパーオキサイドアニオンラジカル）によって生じる過酸化亜硝酸（パーオキシナイトライト）によってRNAがニトロ化された核酸です。
生体組織が炎症を起こすと多量のNOが産生され、多くの過酸化亜硝酸が生じて、グアノシン,デオキシグアノシンをニトロ化することが知られております。
8-Nitroguanosineの局在や量などを調べることにより、8-Nitroguanosine残基の役割や遺伝子傷害、発癌機構の解明に役立つことが期待されます。
＊ポリクローナル抗体で染色された場合、次のような確認を行なうことをお勧めします。
1.標準の8-Nitroguanineと競合させて染色されなくなること
2.Sodium hydrosulfiteなどの還元剤で試料中のニトログアノシン、ニトログアニンをアミノグアノシン、アミノグアニンに還元して染色されなくなること
抗ニトログアノシン抗体はどのくらい保存できますか？
有効期間は設けておりませんが、希釈前の段階で冷蔵で1年使用できることを確認しております。

Specificity Ubi-1 was initially screened on ELISA of the immunogen, and subsequently tested on sections of Alzheimer brain. It was one of several
clones which stained neurofibrillary tangles in frozen sectioned material strongly and specifically. Subsequent studies indicated that Ubi-
1 is relatively insensitive to formalin fixation and so can be used on mildly fixed histological sections of human brain for studies of
Alzheimer's disease. Ubi-1 also recognizes other ubiquitinated inclusion bodies such as the Lewy bodies of Parkinson's disease and
the Pick bodies in Pick's disease. This antibody recognizes polyubiquitin chains much more strongly than monoubiquitinated molecules
or free ubiquitin. Specifically recognizes ubiquitinated cytoplasmic inclusion bodies.

Target
Relevance Ubiquitin is a highly conserved protein of about 8.5 kDa molecular weight, which has an ATP dependent role in the targeting of proteins
for proteolytic degradation. To perform this function, the protein to be degraded is first covalently attached to the C terminus of ubiquitin,
and the ubiquitinated complex is then recognized by a complex of degradative enzymes. Interestingly, ubiquitin also becomes covalently
bonded to many types of pathological inclusions, which appear to be resistant to normal degradation. Therefore, ubiquitin antibodies
are very useful for studies of these inclusions. For example, the neurofibrillary tangles and paired helical filaments diagnostic of
Alzheimer's disease, Lewy bodies seen in Parkinson's disease, and Pick bodies found in Pick's disease are all heavily ubiquitinated
and can be readily visualized with ubiquitin antibodies.

Relevance
Cytokeratins, a group comprising at least 29 different proteins, are characteristic of epithelial and trichocytic cells. Cytokeratins 1, 4, 5, 6, and 8 are members of the type II neutral to basic subfamily. Monoclonal anti cytokeratins are specific markers of epithelial cell differentiation and have been widely used as tools in tumor identification and classification. Monoclonal Anti Pan Cytokeratin （mixture） is a broadly reactive reagent, which recognizes epitopes present in most human epithelial tissues. It facilitates typing of normal, metaplastic and neoplastic cells. Synergy between the various components results in staining amplification. This enables identification of cells, which would otherwise be stained only marginally. The mixture may aid in the discrimination of carcinomas and nonepithelial tumors such as sarcomas, lymphomas and neural tumors. It is also useful in detecting micrometastases in lymph nodes, bone marrow and other tissues and for determining the origin of poorly differentiated tumors. There are two types of cytokeratins the acidic type I cytokeratins and the basic or neutral type II cytokeratins. Cytokeratins are usually found in pairs comprising a type I cytokeratin and a type II cytokeratin. Usually the type II cytokeratins are 8kD larger than their type I counterparts.

ab6401 staining pan Cytokeratin in Mouse liver tumor tissue sections by IHC-P （Paraformaldehyde fixed paraffin embedded sections）. Tissue was fixed with paraformaldehyde and blocked with maleate buffer blocking solution for 30 minutes and 22°C. Antigen retrieval was by heat mediation in citrate buffer. Samples were incubated with primary antibody （1/500） in maleate buffer blocking solution for 16 hours at 22°C. An undiluted biotin-conjugated Donkey polyclonal to mouse IgG was used as secondary antibody.
